【摘要信息】:Gene imprinting is an important phenomenon during mammalian embryo development or growth regulation. In 1983, an inheritable muscular hypertrophy in sheep was reported, and shown to be determined by callipyge gene (CLPG) mapped to ovine chromosome 18 within the DLK1-GTL2 region. This region was corresponded to human imprinted domain located on Chromosome 14q32, including paternal expressed gene DLK1, maternal expressed gene MEG3 (or GTL2), and imprinting status unknown genes DAT, CLBG1, PEG11, MEG8 and DIO3. Previous studies showed that a uniparental disomy of the human chromosomal region was associated with multiple congenital anomalies, including growth retardation and obesity. In addition, the polar overdominant inheritance of a DLK1 polymorphism also influenced growth and fat deposition in the pig. The objectives of this study were to confirm if the porcine DLK1- GTL2 region is imprinted domain as human and sheep and to analyze phenotypic association in a commercial pig population. We have detected two single nucleotide polymorphisms (SNPs) in the coding region of the porcine DLK1 gene, and three SNPs in exon1 of MEG3 gene, and the imprinting detection work is underway. We also designed the primers of MEG8 and PEG11 gene for genomic and cDNA cloning and SNP scanning. The results may increase the knowledge of expression pattern and function of imprinting genes within the DLK1-GTL2 region in mammals.
